Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding mature erythrocytes and platelets. It plays an important role in lymphocyte activation through its tyrosine phosphatase activity.

This antibody has been reported to inhibit cytotoxic activity, activate cell signaling, and deplete NK cells in vivo. It is validated for flow cytometric analysis of mouse bone marrow cells at ≤0.5 µg/test (in 100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration is recommended for best performance.

Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended detection filter: 710/50 bandpass. Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright conjugates to minimize polymer interactions.

Light Sensitivity: Protect vial and samples from light to prevent photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal effect on brightness and FRET efficiency.

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, constituting ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic cells. CD45 regulates Src-family kinases, T/B-cell antigen receptor signaling, and cytokine receptor pathways. Isoforms generated by alternative splicing differ in extracellular domains and expression patterns. CD45 also modulates integrin-mediated signaling, apoptosis, and various immunological functions.
