Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed either as a heterodimer with CD8 beta (CD8α/β) or as a homodimer (CD8α/α). CD8a is present on most thymocytes and subsets of mature T cells and NK cells. CD8 binds to MHC class I and, through association with p56lck, participates in T-cell development and activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
• NovaFluor conjugates use Phiton™ technology based on nucleic acid dye structures for engineered fluorescence with minimal spillover.

Target Information

CD8 (Cluster of Differentiation 8):
A type I transmembrane glycoprotein receptor of the immunoglobulin family involved in T-cell signal transduction, differentiation, and activation.

• Expressed primarily on cytotoxic T lymphocytes as CD8α/β heterodimers.
• Also found as CD8α/α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
• Functions as a co-receptor for MHC class I molecules.
• Engagement of MHC-I/peptide complexes triggers Lck recruitment, lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
• Differentiation of CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
