Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein. CD21 is expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells and acts as a receptor for complement component C3d and Epstein-Barr virus (EBV). CD21, together with CD19 and CD81, forms a multimolecular complex with BCR and is involved in B cell activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.05 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation for optimal staining. Add 5 µL per sample to antibody cocktails prior to cell addition. For single-color controls, use 5 µL per 100 µL sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures designed to minimize spillover and spread effects.

Target Information

CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor) binds C3 complement fragments, facilitating immune complex uptake and memory B cell survival. It plays a role in humoral immune response development and serves as a receptor for Epstein-Barr virus and prions. Soluble CD21 retains ligand-binding ability. CD21 associates with CD19, CD81, and Leu13, forming a signal-transduction complex involved in B cell activation. Genetic variations in CD21 are linked to susceptibility to systemic lupus erythematosus type 9 (SLEB9). Alternative splicing produces multiple isoforms.
