Target Information
Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
FPGS plays a crucial role in folate homeostasis in mammals by:
- Preventing efflux of folate cofactors from cells via polyglutamation
- Enhancing binding of folate cofactors to enzymes involved in folate interconversion and biosynthesis
- Facilitating accumulation of folates in mitochondria for glycine synthesis
FPGS is widely expressed in human tumors and is associated with cell proliferation or lack of differentiation. Its integrative function increases the efficiency of folate antimetabolites used in cytotoxic therapies.
