Recommended positive control tissue: ZMTB-5 (other tissues may also be suitable).

The primary antibody is intended for professional laboratory use in detecting CD56 protein in formalin-fixed, paraffin-embedded tissue for manual qualitative IHC testing. It is used after tumor diagnosis by conventional histopathology.

Three isoforms of NCAM (CD56) are produced by alternative splicing. The 135 kDa isoform is glycosylated or sialylated to form mature species. NCAM is expressed on neuroectodermal-derived cell lines and neoplasms such as retinoblastoma, medulloblastoma, astrocytoma, and neuroblastoma, as well as mesodermal tumors like rhabdomyosarcoma and natural killer cells.

Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, followed by washing steps. The resulting chromogenic reaction is visualized under a light microscope.

A positive tissue control must be included in each staining procedure. This control should contain both positive and negative staining components. External positive controls should be fresh, properly processed specimens fixed and embedded similarly to patient samples. Tissues with weak positive staining are preferred for detecting subtle changes in antibody sensitivity or reagent degradation.

Negative control tissue (internal or external) should confirm absence of specific staining and indicate background levels. If specific staining occurs in negative controls, patient specimen results are invalid.

Target Information

CD56 (NCAM, neural cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin family involved in cell adhesion, migration, axonal growth, and synaptic plasticity. It is expressed in the nervous system and on T and NK cells, existing in isoforms of 120–180 kDa. Polysialic modification reduces adhesion. CD56 mediates homophilic and heterophilic interactions with extracellular matrix components like laminin and integrins. It is widely used as a neuroendocrine marker for neuroendocrine and ovarian granulosa cell tumors. CD56 dysfunction is associated with rabies and blastic plasmacytoid dendritic cell neoplasms.
