Target Information
Cardiac Troponin I (TNNI3) is an integral inhibitory protein in cardiac muscle, forming a complex with troponin C (TnC) and troponin T (TnT). The TnT subunit binds to tropomyosin, creating a troponin–tropomyosin complex anchored by TnI binding to actin within muscle thin filaments.
Calcium binding to TnC induces a structural change that releases the inhibitory region of TnI from actin, enabling myosin attachment and muscle contraction.
Blood levels of TnI and TnT are important diagnostic indicators of cardiac muscle damage, useful for distinguishing angina from myocardial infarction.
Cardiac Troponin I also inhibits angiogenesis in vivo and in vitro. Mutations in the TNNI3 gene are associated with familial hypertrophic cardiomyopathy type 7 (CMH7) and familial restrictive cardiomyopathy (RCM).
