Target Information
The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes and required for RNA polymerase II transcription. MED28 (also known as Magicin) is one of the mediator subunits, expressed in various cell lines and tissues.
Down-regulation of MED28 in NIH3T3 cells induces genes related to smooth muscle cell differentiation, while overexpression represses SMC gene expression.
MED28 can form a ternary complex with Grb2 and Merlin (NF2 tumor suppressor protein), suggesting a role in tumor suppression.
It has also been identified as an HIV dependency factor (HDF), implying potential as a drug target in HIV research.
At least two isoforms of MED28 are known to exist.
