Target Information
The structure-specific ERCC1/XPF endonuclease complex is involved in repairing two distinct types of DNA lesions:
- Nucleotide excision repair (NER) for UV-induced lesions and bulky chemical adducts
- Recombination repair of genotoxic interstrand cross-links
In NER, dual incisions occur on both sides of the DNA damage, catalyzed by two nucleases. In mammalian cells, XPG cleaves the 3′ side of the lesion, while the ERCC1/XPF complex makes the 5′ incision. ERCC1/XPF also likely participates in processing recombination intermediates.
