Target Information
DMPK는 다양한 근육 및 신경 조직에서 발현되는 세린/트레오닌 키나아제입니다. DMPK 유전자의 비번역 영역 돌연변이는 근긴장성 이영양증의 특징으로 알려져 있습니다. 이 단백질은 Rho 계열의 작은 GTPase와 상호작용하는 다른 키나아제와 밀접하게 관련되어 있으며, 기질로는 myogenin, L-type 칼슘 채널의 베타 서브유닛, phospholemman 등이 있습니다.
3’ 비번역 영역에는 CTG 삼염기 반복 서열(5–37 copies)이 존재하며, 이 반복이 50–5,000 copies로 확장되면 근긴장성 이영양증 I형을 유발하며 반복 수가 증가할수록 질병의 중증도가 높아집니다. 반복 확장은 국소 염색질 구조의 응축을 유발하여 해당 영역의 유전자 발현을 방해합니다.
