Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms of Ki-67 exist (345 and 395 kDa) expressed in dividing cells. Ki-67 is present in all cell types during active cell cycle phases (G1, S, G2, and mitosis) but absent in resting cells (G0). During interphase, Ki-67 is localized to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.

In tumor studies, Ki-67 expression serves as a marker for the fraction of proliferating cells. This antibody also recognizes canine Ki-67.

Applications Reported: Suitable for immunocytochemical and immunohistochemical staining of paraffin-embedded tissue (IHC-P).

Applications Tested: Validated in immunocytochemistry on formaldehyde-fixed and permeabilized HeLa cells, and immunohistochemistry on FFPE human skin using low pH antigen retrieval. Recommended use ≤1 µg/mL; titrate for optimal performance.

Filter Recommendation: Use filters capturing 615 nm emission wavelength (standard Alexa Fluor® 594 filter acceptable).

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain that mediates interactions with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus. Ki-67 shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.

Ki-67 is widely used as a proliferation marker since it is absent in G0 cells and increases through mitosis. It helps determine the growth fraction in neoplasms, correlating with tumor grade and prognosis. Commonly used as a neuronal cell proliferation marker.
