Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator. The 20S core has four rings made of 28 non-identical subunits (two alpha and two beta rings). The 19S regulator consists of a base with ATPase and non-ATPase subunits and a lid with up to 10 non-ATPase subunits. Proteasomes are abundant in eukaryotic cells and function in ATP/ubiquitin-dependent peptide cleavage. The immunoproteasome, a modified form, processes class I MHC peptides and contains the 11S regulator (PA28) instead of the 19S. The gamma subunit of the 11S regulator forms a homohexameric ring, and two transcript variants encoding different isoforms have been identified.
