Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin belonging to the Ig superfamily that binds glycoconjugates containing sialic acids. Siglec F is expressed mainly on eosinophils and alveolar macrophages, and at lower levels on immature myelomonocytic cells. Mouse Siglec F serves as a functional ortholog of human Siglec 8 but is not expressed on mast cells. This antibody recognizes a formaldehyde-fixed epitope.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated using mouse thioglycolate-elicited peritoneal exudate cells at ≤0.05 µg/test (100 µL final volume). Cell numbers may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Important Usage Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic-acid dependent binding. It binds both α2,3- and α2,6-linked sialic acids. Expression occurs in monocytic/myeloid lineage cells, including monocytes and neutrophils, and at high levels in peripheral blood leukocytes, spleen, and bone marrow.
