Target Information
L-glutamate는 포유류 중추신경계의 주요 흥분성 신경전달물질입니다. 흥분독성 수준을 방지하기 위해, 시냅스 간극의 과잉 glutamate는 이온 결합성 glutamate 수송체에 의해 제거됩니다. 주요 수송체로는 GLAST-1, GLT-1, EAAC-1, EAAT-4가 있으며, EAAC-1은 중추신경계뿐 아니라 신장과 소장에서도 발현됩니다.
EAAC-1은 8개의 막관통 α-helix와 독특한 loop-pore 구조를 가지며, 이 구조는 이온 채널의 loop-pore와 유사합니다. 또한, 막을 통과하는 친수성 경로를 제공하는 고도로 양친성 막관통 helix를 포함합니다.
EAAC-1 유전자 결손 마우스는 dicarboxylic aminoaciduria를 나타내며, 자발적 운동 활동이 감소하지만 12개월 동안 신경퇴행은 관찰되지 않았습니다.
