Target Information
FAM21A functions as a component of the WASH core complex, regulating actin polymerization at endosomal surfaces. It mediates recruitment of the WASH complex to endosome membranes via binding to phospholipids and the retromer CSC subunit VPS35. This process is essential for vesicle fission and endosomal sorting. FAM21A also participates in endosome-to-plasma membrane trafficking and recycling of SNX27-retromer-dependent cargo proteins such as GLUT1. It is required for association of DNAJC13, ENTR1, ANKRD50 with VPS35, and for recruitment of CCC and retriever complex subunits (COMMD1, CCDC93, VPS35L).
