Target Information
Nalcn is a voltage-independent, cation-nonselective channel permeable to sodium, potassium, and calcium ions. It regulates the resting membrane potential and neuronal excitability (PubMed:17448995). Neuropeptides such as neurotensin and substance P stimulate action potentials by activating NALCN via a SRC family kinase-dependent pathway. Nalcn activity is modulated by GPCRs, essential for normal respiratory rhythm, neonatal survival, and systemic osmoregulation through serum sodium control. It contributes to substance P-induced depolarization and intestinal pacemaker regulation, and plays a critical role in spontaneous firing and excitability modulation of substantia nigra pars reticulata (SNr) neurons.
Diseases associated with NALCN include congenital contractures of the limbs and face, hypotonia, developmental delay, and infantile psychomotor retardation with characteristic facies.
