Target Information
FXR (Farnesoid X receptor; NR1H4)는 ligand-activated transcription factor로 작용하는 nuclear receptor superfamily (NR1 subfamily)에 속합니다. FXR은 chenodeoxycholic acid, lithocholic acid, deoxycholic acid 등의 담즙산을 리간드로 인식하며, 담즙산 합성, 결합 및 장간순환에 관련된 유전자 조절을 통해 담즙산 항상성 유지에 중요한 역할을 합니다. 또한 지질 및 포도당 항상성 조절, 선천 면역 반응에도 관여합니다.
FXR-RXR heterodimer는 FXRE(farnesoid X receptor response elements)에 결합하며, 5’-AGGTCA-3’ 서열을 가진 inverted repeat(IR-1) 구조에 높은 친화도로 결합합니다. 간에서 FXR은 NR0B2(corepressor)를 활성화하여 CYP7A1 및 CYP8B1의 발현을 억제하고, SLC10A1/NTCP, SLC27A5/BACS, BAAT, ABCB11/BSEP, ABCC2/MRP2, ABCB4 등의 유전자를 조절하여 담즙산 및 인지질 대사에 관여합니다.
