SIRT5 is a member of the sirtuin (Sir2-like) protein family found in both prokaryotes and eukaryotes.
All sirtuins share a conserved core domain that includes motifs from bacteria to humans.
Sirtuins utilize NAD⁺ and may function as mono-ADP-ribosyltransferases.
Although the complete enzymatic role of sirtuins remains under investigation, studies suggest a link between NAD metabolism and histone deacetylation, indicating a possible reciprocal activation mechanism involving Sir2, NAD, and acetylated histones.
