Target Information
FOXO1 (FKHR, Forkhead Box O1)는 전사 인자(forkhead homeotic gene family)의 하위 그룹 구성원으로, 핵 수용체 활성을 조절하는 공활성자(coactivator) 또는 공억제자(corepressor)로 작용할 수 있습니다.
FOXO1은 PAX3와의 결합을 통해 횡문근육육종(alveolar rhabdomyosarcoma)과 관련이 있으며, 자가포식(autophagy), 근육 성장 및 분화, 항종양 활성 등 다양한 생리적 과정에 관여합니다.
또한 인슐린 신호전달의 주요 표적으로서 산화 스트레스에 대한 대사 항상성을 조절하며, IRE(Insulin Response Element) 및 DBE(Daf-16 Binding Element)에 결합합니다.
FOXO1은 골 형성 조절, 적색 산화 균형 유지, 골격의 내분비 기능 조절, 그리고 RUNX2 억제를 통한 오스테오칼신 발현 조절에도 중요한 역할을 합니다.
간세포에서는 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PCK1 등의 유전자 발현을 활성화함으로써 포도당신생합성을 촉진합니다.
FOXO1은 CDK1, PKB/AKT1, MST1 신호 하류에서 세포 사멸을 조절하는 핵심 인자입니다.
