Target Information

항체의 동형(Isotype)과 사용되는 응용 분야에 따라 배경 염색이 발생할 수 있습니다. 이러한 비특이적 신호는 Fc 수용체 결합, 세포 내 단백질·탄수화물·지질과의 비특이적 상호작용, 또는 세포 자가형광에 의해 유발됩니다.
Isotype control 항체는 특정 항원에 대한 특이성이 없으므로, 비특이적 배경 신호와 실제 항체 신호를 구분하는 음성 대조군으로 활용됩니다.
Flow Cytometry에서 가장 흔히 사용되지만, ChIP, Immunohistochemistry, Gel Shift 등 다양한 응용 분야에서도 유용하게 사용됩니다.
Isotype control은 주항체(primary antibody)의 종(species)과 동형(isotype)을 일치시켜야 하며, 직접 표지된 항체를 사용할 경우 동일한 형광 라벨로 결합된 대조군을 사용하는 것이 가장 적합합니다.
