EPHA3는 단백질 티로신 키나아제 계열에 속하는 ephrin 수용체 하위군에 속합니다. EPH 및 관련 수용체들은 특히 신경계 발달 과정에서 중요한 역할을 하는 것으로 알려져 있습니다.
EPHA3는 ephrin-A 리간드와 결합하며, EPH 수용체 하위군의 단백질들은 일반적으로 하나의 키나아제 도메인과 Cys-rich 도메인 및 두 개의 fibronectin type III 반복을 포함하는 세포외 영역을 가집니다.
Ephrin 수용체는 세포외 도메인 서열의 유사성과 ephrin-A 및 ephrin-B 리간드에 대한 결합 친화도에 따라 두 그룹으로 나뉘며, EPHA3는 ephrin-A 리간드에 결합합니다.
