Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also facilitates HIV-2 binding via envelope glycoprotein gp120, promoting Env-mediated viral fusion. CXCR4 serves as the receptor for chemokine CXCL12/SDF-1, mediating intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling. CXCR4 plays roles in hematopoiesis, cardiac septum formation, and gastrointestinal vascularization, and is involved in neuronal survival and cerebellar development. It binds bacterial LPS, mediating inflammatory responses such as TNF secretion by monocytes. Antibodies targeting CXCR4 can block HIV-1 and HIV-2 fusion and infection of human cells.
