Description

These kits provide a fast, reliable, and convenient method to sequentially isolate and concentrate exosomal RNA and free-circulating RNA from different urine sample volumes.
The purification is based on spin column chromatography employing Norgen’s proprietary resin.
All sizes of RNA, including microRNA, are isolated without bias and without special instrumentation or toxic reagents.
The purified RNA is suitable for downstream applications such as:

  • Real-time PCR
  • Reverse transcription PCR
  • Northern blotting
  • RNase protection and primer extension
  • Expression array assays

Storage Conditions and Product Stability

All buffers should be tightly sealed and stored at room temperature.
Kit stability: 2 years after shipment.
If salt precipitation is observed, warm Lysis Buffer A for 20 minutes at 60°C.


Important Note

Urine samples stored at -70°C, -20°C, or 4°C may develop precipitation due to protein aggregation, which can cause exosome loss and affect RNA quality.
Use Norgen’s Urine Preservative for optimal nucleic acid and protein preservation at ambient temperature.
Samples can be stored for over 2 years without degradation.

Norgen’s Urine Exosome and Free-Circulating RNA Isolation Midi Kit (Cat# 59300) was used to isolate exosomal RNA from urine volumes ranging from 2 mL to 10 mL. Two microlitres of isolated RNA were used in RT-qPCR to assess urinary exosomal miR-30a amplification.
(A) The urinary exosomal miR-30a decreases linearly with increasing sample input volume.
(B) Relative amount shows excellent linearity with >90% recovery.

Using the same kit, free-circulating RNA was isolated following exosomal RNA purification from urine volumes of 2–10 mL. Two microlitres of RNA were used in RT-qPCR for urinary free-circulating miR-21 amplification.

(B) Relative amount shows excellent linearity with >90% recovery.
