Target Information
SPT6는 전사 연장 인자로서 히스톤 H3에 결합하고 전사 연장 및 mRNA 가공 조절에 중요한 역할을 합니다. RNA polymerase II (RNAPII)의 전사 연장을 촉진하며, HIV-1 Tat에 의한 전사 활성화에도 필요합니다. 또한 IWS1 및 RNAPII의 RPB1 서브유닛의 C-말단 도메인(CTD)과 복합체를 형성하여 mRNA 수송 및 스플라이싱을 조절합니다. 이 복합체는 mRNA export 인자(ALYREF/THOC4, EXOSC10)와 히스톤 변형 효소(SETD2 등)를 모집하여 효율적인 mRNA 수출 및 H3K36 메틸화(활성 전사의 표지)를 유도합니다. SUPT6H는 SETD1A와의 상호작용을 통해 AICDA 표적 유전자에서 H3K4me3 표지를 형성하여 class-switch 재조합 및 체세포 돌연변이를 조절합니다. 또한 KDM6A의 안정화를 통해 근육 형성 유전자 프로그램을 활성화합니다.
