Target Information
TRAIL은 종양괴사인자(TNF) 리간드 패밀리에 속하는 사이토카인으로, 변형된 종양세포에서 선택적으로 세포사멸(apoptosis)을 유도하지만 정상 세포에는 영향을 거의 미치지 않습니다.
TRAIL은 TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, TNFRSF11B/OPG 등 다양한 TNF 수용체 슈퍼패밀리 구성원에 결합합니다.
활성은 apoptosis를 유도하지 않는 decoy receptor인 TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, TNFRSF11B/OPG에 의해 조절될 수 있습니다.
TRAIL 수용체 결합은 MAPK8/JNK, caspase 8, caspase 3의 활성화를 유도하며, 두 개의 death receptor (DR4, DR5)와 두 개의 decoy receptor (DcR1, DcR2)를 포함합니다.
TRAIL은 다양한 인체 조직에서 발현되는 type II 막 단백질로, TNF 및 Fas ligand와 유사하게 apoptosis와 NF-κB 활성화를 유도합니다.
