Target Information

Oxygen radicals damage chromosomal DNA, causing cell death and inducing mutations. Although much attention has focused on repairing 8-oxo-G lesions, various other DNA damages are also caused by free-radical attack. The human NTH1 repair protein acts on multiple oxidatively damaged DNA sites. As a homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. It exhibits broad substrate specificity, recognizing numerous ring saturation and fragmentation products of pyrimidines. Research indicates that NTH plays a crucial role in the removal of oxidative base lesions in mitochondrial DNA.
