Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins linking the TNF receptor superfamily to signaling pathways, regulating cell death and cellular stress responses.
TRAF proteins share a homology region that enables binding to cell receptors and other TRAF proteins, activating distinct signaling cascades.
TRAF2 and TRAF3 directly bind to CD40, a NF receptor superfamily member involved in B cell immunity, and are critical for NF-kappa-B activation in mouse B lymphocytes.
TRAF2, along with TRAF6, is required for CD40 signaling in nonhemopoietic cells and interacts with LTbetaR in association with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
