Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor. CD86 and CD80 are members of the B7 family of costimulatory molecules.
CD86 is expressed on B cells, macrophages, and dendritic cells and upregulated through stimuli such as BCR complex, CD40, and cytokine receptors.
It interacts with CD28 and CD152 (CTLA-4) on T cells, playing a key role in T-B crosstalk, T cell costimulation, and antibody production.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested with normal human peripheral blood cells at 5 µL (0.2 µg)/test.
Cell number range: 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

Blocking Buffer Recommendation:
Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for all experiments with NovaFluor conjugates.
Add 5 µL per sample before combining with cells or antibody cocktails.

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation.
It is expressed by antigen-presenting cells and regulates immune responses through its interactions with T-cell surface proteins.
Alternative splicing produces multiple isoforms.
Associated diseases include gallbladder squamous cell carcinoma and myocarditis.
