Target Information
Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca(2+)/calmodulin-binding and autophosphorylation. Involved in dendritic spine and synapse formation, neuronal plasticity, and regulation of sarcoplasmic reticulum Ca(2+) transport in skeletal muscle.
In neurons, it plays an essential structural role in actin cytoskeleton reorganization during plasticity by binding and bundling actin filaments in a kinase-independent manner. This function is required for correct targeting of CaMK2A, promoting dendritic spine and synapse formation and maintaining synaptic plasticity enabling long-term potentiation (LTP) and hippocampus-dependent learning.
In developing hippocampal neurons, it promotes dendritic arborization, and in mature neurons, dendritic remodeling. It also modulates skeletal muscle function in response to exercise, regulating SR Ca(2+) transport in slow-twitch muscle and controlling Ca(2+) release in fast-twitch muscle via phosphorylation of triadin and phospholamban.
