Cellular Localization	Cytoplasm; Cell membrane (peripheral membrane protein); Nucleus. In steady state, BTK is predominantly cytosolic. Following B-cell receptor (BCR) engagement, it translocates to the plasma membrane via its PH domain. A fraction also shuttles between the nucleus and cytoplasm, with export mediated by CRM1.
Background	The BTK gene encodes a protein crucial for B-cell development. Mutations cause X-linked agammaglobulinemia type 1, an immunodeficiency characterized by failure to produce mature B lymphocytes and defective Ig heavy chain rearrangement.
