Target Information
Proteolytic degradation is critical for maintaining appropriate levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins, while others are degraded via the vacuolar pathway (endosomes, lysosomes, ER).
The 26S proteasome is an ATP-dependent, multisubunit (~31), barrel-shaped molecular machine (~2.5 MDa) consisting of a 20S proteolytic core complex with 19S regulatory subunits that recognize ubiquitinated proteins and unfold/translocate them.
Defects in this pathway are linked to genetic diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
