Target Information
NLK is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway, downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation results in phosphorylation of SETDB1, forming the NLK-SETDB1 complex that interacts with PPARG to promote histone H3K9 methylation and transcriptional silencing. This inhibits adipogenesis and promotes osteoblastogenesis in mesenchymal stem cells. NLK also negatively regulates the canonical Wnt/β-catenin pathway by phosphorylating TCF7L2/TCF4 and LEF1, disrupting the β-catenin complex and promoting LEF1 degradation. Additionally, NLK inhibits Notch signaling by phosphorylating NOTCH1, preventing formation of the active ternary complex with RBPJ/RBPSUH and MAML1. It also negatively regulates MYB family transcription factors.
