Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and acts as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression, as well as cytotoxicity of CD3-activated T cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended bandpass filter: 660/20. Confirm instrument compatibility before use.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal loss of brightness.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and serves as a critical co-stimulatory receptor, binding CD80 and CD86 on antigen-presenting cells. CD28 prevents T cells from entering an anergic or apoptotic state.

In murine lymphoid organs, CD28 is expressed on all CD4+ and CD8+ T cells. Expression is highest on immature thymocytes and decreases in mature populations. CD28 dysfunction is associated with diseases such as mycosis fungoides and Sezary’s disease.
