Target Information
PUMA is a pro-apoptotic member of the Bcl-2 family, including Bax and Noxa, which are transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins, PUMA-a and PUMA-b. PUMA proteins bind Bcl-2, localize to mitochondria, and induce cytochrome c release and apoptosis in response to p53 activation. PUMA may act as a direct mediator of p53-induced apoptosis. Inhibition of PUMA expression reduces the apoptotic response to p53, indicating its role in cytochrome c/Apaf-1-dependent cell death pathways.
