Target Information

HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is membrane-anchored and plays a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. These molecules are expressed in nearly all cells.

The heavy chain (~45 kDa) gene contains 8 exons:

  • Exon 1: Leader peptide
  • Exons 2–3: Alpha1 and Alpha2 domains (peptide binding)
  • Exon 4: Alpha3 domain
  • Exon 5: Transmembrane region
  • Exons 6–7: Cytoplasmic tail

Polymorphisms within exons 2 and 3 determine the peptide-binding specificity of each class I molecule. Typing for these polymorphisms is routinely performed for bone marrow and kidney transplantation. Numerous HLA-A alleles have been described.

HLA and MHC antibodies are important in Immunopeptidomics, aiding in the identification and characterization of neoantigens via high-performance liquid chromatography coupled with tandem mass spectrometry.
