Target Information
CYLD는 956개의 아미노산으로 구성된 세포질 내 탈유비퀴틴화 효소로, ubiquitin carboxy-terminal hydrolases (UCH) 단백질 계열에 속합니다. 이 단백질은 세 개의 CAP-GLY 도메인, proline-rich 영역, SH3 결합 도메인, 그리고 UCH 촉매 도메인과 유사한 서열을 포함합니다.
CYLD는 JNK 신호전달 경로에 영향을 미치는 종양 억제 단백질로 알려져 있으며, TRAF2 및 NF-κB 신호전달 경로의 음성 조절자로 작용합니다. 또한 I-κB kinase 경로 조절에 수용체 의존적 역할을 가지며, Lys-48 비연결 폴리유비퀴틴 사슬에 대한 탈유비퀴틴화 활성을 갖습니다. TRAP1은 CYLD의 새로운 탈유비퀴틴화 기질로 보고되었습니다.
CYLD 유전자의 돌연변이는 cylindromatosis, multiple familial trichoepithelioma, Brooke-Spiegler syndrome과 관련이 있습니다.
