Target Information

Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase acting as a substrate for Src and a key component of integrin signaling. It plays crucial roles in cell spreading, differentiation, migration, apoptosis, and cell cycle progression (G1 to S phase transition).
FAK localizes to focal adhesions, mediating cell attachment to the extracellular matrix via integrins. Tyrosine phosphorylation of FAK increases upon integrin engagement with fibronectin. Adhesion of NIH3T3 fibroblasts to fibronectin promotes association of Grb2 and c-Src with FAK, activating ERK2 MAP kinase. In v-Src-transformed NIH3T3 cells, association of v-Src, Grb2, and Sos with FAK occurs independently of fibronectin adhesion.
The Grb2 SH2 domain binds directly to tyrosine-phosphorylated FAK, with Tyr925 identified as the binding site. Multiple transcript variants exist for FAK, with three full-length isoforms characterized.
