Target Information
Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that allows association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows a cell cycle-dependent distribution:
- G1 phase: perinucleolar expression
- G2 phase: nuclear matrix expression
- M phase: chromosomal expression
As Ki-67 is absent in G0 cells but increases through mitosis, it is widely used as a proliferation marker to determine the growth fraction of neoplasms. The Ki-67 index correlates strongly with tumor grade and is a valuable prognostic indicator. It is also used as a neuronal marker for cell cycling and proliferation.
