Target Information
The PKC family of serine/threonine kinases, including PRKCI (PKC iota), is activated intracellularly by signal transduction pathways. In humans, at least 12 different PKC polypeptides have been identified, differing in primary structure, tissue distribution, subcellular localization, mode of action, and substrate specificity. PKC alpha, beta I, beta II, and gamma form the conventional family, which are Ca²⁺- and phospholipid-dependent.
PKC iota is a calcium-independent, phospholipid-dependent kinase not activated by phorbol esters or diacylglycerol. It interacts with the small GTPase RAB2, phosphorylating GAPDH and contributing to microtubule dynamics in the early secretory pathway. PKC iota is required for BCL-ABL-mediated resistance to drug-induced apoptosis, thus protecting leukemia cells. A single exon pseudogene is mapped on chromosome X.
