VEGFA (Vascular Endothelial Growth Factor A), also known as Vascular Permeability Factor (VPF), is a major signaling growth factor belonging to the VEGF family, a subgroup of the Platelet-Derived Growth Factor (PDGF) proteins characterized by a conserved cystine-knot structure. VEGF expression is upregulated under hypoxia and in many types of cancers. VEGF receptors (VEGFR1 and VEGFR2) are often overexpressed in tumors, and high VEGF levels correlate with poor prognosis. Inhibition of VEGF-mediated angiogenesis pathways has potential in angiogenesis-related disease therapy, achievable by blocking circulating VEGF with antibodies such as Bevacizumab or Aflibercept, or by VEGFR-2 inhibition using small molecules like Gamabufotalin (CS-6) and Arenobufagin.
