Bridging integrator-1 (BIN1), also called amphiphysin II, is a nucleocytoplasmic protein that is ubiquitously expressed. It binds to the transcription factor Myc and interacts with SRC SH3, a tyrosine kinase ligand. Amphiphysin II belongs to the amphiphysin family, along with amphiphysin I.
The human BIN1 gene is located on chromosome 2q14 and contains 20 exons. Alternative splicing produces multiple isoforms: isoform 8 in skeletal muscle, 1–7 in brain, and 9–10 expressed ubiquitously. BIN1 (also known as SH3P9) includes an N-BAR domain, phosphoinositide-binding motif, clathrin and AP2 binding domain, Myc-binding domain, and SRC homology 3 domain.
