The gene APOA4 is localized to the long arm of human chromosome 11 along with A1 (APOA1) and C3 (APOC3) genes. This gene contains three exons separated by two introns. Apo A-IV is synthesized in the mucosal cells of the small intestine as a preprotein. This 396-amino acid preprotein undergoes proteolytic processing, associates with chylomicrons during fat absorption, and is secreted into the lymph.
