It contains a constitutive promoter (OXB20) derived from the E. coli RecA region, requiring no induction or additional components for activity.
OXB20 is the strongest bacterial promoter offered by Merck Sigma, providing high expression levels that may cause solubility issues for certain proteins.
Alternative promoters with lower expression levels (OXB1–OXB20) are available upon request.

About the Cleavage Tag

The plasmid encodes a Thrombin protease cleavage site positioned between the gene of interest and the HA tag, allowing tag removal post-purification.

Thrombin cleaves preferentially between Arg and Gly residues. Off-target cleavage may occur due to contaminating proteases; thus, high-purity enzyme reagents are recommended.

Promoter Expression Level

The OXB20 promoter provides strong constitutive expression in E. coli.
For proteins with poor solubility, alternative promoter variants with lower expression strength are available.
