Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, resulting in G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is linked to tumor formation. p53 plays roles in tumor suppression, aging, oxidative stress response, and neuronal plasticity. Mutations in TP53 often lead to loss of tumor suppressor activity and are associated with various cancers and syndromes such as Li-Fraumeni.
