Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating p53 stability. It is located on chromosome 12q and is phosphorylated by Akt at Ser166 and Ser186, enhancing its interaction with p300 and promoting p53 ubiquitination and degradation. Phosphorylation also prevents MDM2 from binding to p19ARF, further increasing p53 degradation. Overexpression of MDM2 leads to p53 inactivation and reduced tumor suppressor function. MDM2 also interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, influencing apoptosis, cell cycle, and tumorigenesis. It is overexpressed in various human malignancies, including soft tissue carcinomas and breast cancer. More than 40 alternatively spliced transcript variants have been identified in tumor and normal tissues.
