Product Specific Information
For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional round of desalting using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
Modification of target proteins by ubiquitin participates in diverse biological functions. Proteins destined for degradation or processing via the 26S proteasome are coupled to multiple copies of ubiquitin. Attachment of ubiquitin or related molecules can alter subcellular distribution or protein activity.
Deubiquitination, the removal of ubiquitin, is catalyzed by deubiquitinating enzymes (DUBs) in four major families:
- Ubiquitin C-terminal hydrolases
- Ubiquitin-specific processing proteases (USPs)
- OTU-domain ubiquitin-aldehyde-binding proteins
- Jab1/Pad1/MPN-domain-containing metallo-enzymes
Among these, USPs are the most widespread. They release ubiquitin from conjugated proteins and contain conserved catalytic domains with Cys and His boxes, while variable N- or C-terminal extensions assist in substrate recognition and localization.
