Target Information
Humans with lipodystrophy exhibit loss of body fat, fatty liver, hypertriglyceridemia, and insulin resistance. Mice with mutations in the fatty liver dystrophy gene show similar phenotypes. Through positional cloning, the mouse gene responsible for fatty liver dystrophy was identified as Lpin1, encoding a nuclear protein named lipin. Lpin1 mRNA is highly expressed in adipose tissue and induced during preadipocyte differentiation, indicating its role in normal adipose tissue development and as a candidate gene for human lipodystrophy.
Database searches revealed homologous sequences in mouse and human genomes: LPIN1, LPIN2, and LPIN3.
