The Human MMP-12 Simplex ProcartaPlex™ Kit measures MMP-12 protein and is designed to be combinable with other Simplex kits, enabling creation of custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation.
When combining multiple Simplex kits (not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously on a Luminex instrument.
These multiplex assays require as little as:

• 25 μL of plasma or serum, or
• 50 μL of cell culture supernatant
  and yield analyzed results in approximately 4 hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample.

작동 원리

Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create unique spectral signatures identifiable by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).
Similar to a sandwich ELISA, each bead is labeled with antibodies specific to a single target protein. Bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).
The conjugation of protein-specific antibodies to distinct beads allows simultaneous analysis of multiple targets within a single well.

ELISA와의 차이점

In ProcartaPlex assays, the capture antibody is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating.
Detection is achieved using dual lasers in Luminex instruments—one distinguishing bead signatures and the other quantifying RPE fluorescence proportional to protein concentration.
This enables profiling of more target proteins using less sample and similar assay time compared to traditional ELISA.
