Target Information
Cell cycle progression is subject to arrest at G1 and G2 checkpoints in response to DNA damage, allowing time for DNA repair before entry into S and M phase. The p53 tumor suppressor regulates one such G1 checkpoint by upregulating GADD45 and p21.
GADD45 binds Cdks and PCNA, proteins involved in DNA replication and repair, and stimulates DNA excision repair while inhibiting S phase entry.
GADD45-like proteins (GADD45β and GADD45γ) are induced by environmental stresses and are thought to activate p38/JNK signaling via MEKK4 activation.
