Target Information
MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases and contains N-terminal catalytic and C-terminal regulatory domains. MAP4K3 acts upstream of MEKK1 and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway. It can directly activate PKC-T during TCR signaling. Enhanced MAP4K3 expression and activation of PKC-T and IKK have been observed in T cells of individuals with systemic lupus erythematosus. MAP4K3 functions as a pro-apoptotic kinase, orchestrating activation of BAX via modulation of PUMA, BAD, and BIM. The kinase activity can be stimulated by UV radiation and TNF-alpha, specifically activating the c-Jun N-terminal kinase (JNK) pathway. Formerly known as RAB8-interacting protein-like 1 (RAB8IPL1), MAP4K3 is now referred to as mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
