Target Information

Levomethadyl acetate is found in individuals who have used or taken this drug. It is a narcotic analgesic with long onset and duration of action, mainly used in the treatment of narcotic dependence.
Opiate receptors (Mu, Kappa, Delta) are G-protein–coupled receptors regulating synaptic transmission via effector proteins. Binding of the opiate stimulates the exchange of GTP for GDP on the G-protein complex, inhibiting adenylate cyclase and decreasing intracellular cAMP. This leads to reduced release of nociceptive neurotransmitters such as substance P, GABA, dopamine, acetylcholine, and noradrenaline. Opioids also inhibit release of vasopressin, somatostatin, insulin, and glucagon. Levomethadyl acetate activates calcium-dependent inwardly rectifying potassium channels (OP1 receptor agonist), causing hyperpolarization and reduced neuronal excitability.
